目的:研究局部使用磁性纳米铁热疗对大鼠胶质瘤的治疗效果。方法:将传代培养的C6细胞立体定向接种于SD大鼠的尾状核,14 d后肿瘤生长至直径0.5~1.0 cm时随机分成对照组和3、5、8 mg磁性纳米铁热疗组,分别立体定向注入等体积的生理盐水及3、5、8 mg的磁性纳米铁,于交变磁场中连续热疗3次,每次30 min,观察大鼠的生存时间及肿瘤体积变化。免疫组织化学SP法检测bcl-2和bax蛋白表达。结果:肿瘤热疗3次后3、5、8 mg磁性纳米铁热疗组大鼠的生存时间分别为(19.60±1.43)、(18.90±1.75)、(20.40±2.26)d,明显长于对照组[(13.30±1.69)d,P〈0.05],各剂量磁性纳米铁热疗组bax蛋白表达亦明显强于对照组,差异有统计学意义(均P〈0.05);各剂量磁性纳米铁热疗组间bax蛋白表达、bcl-2蛋白表达及生存时间比较差异无统计学意义(均P〉0.05)。结论:磁性纳米铁热疗能有效地促进大鼠胶质瘤细胞凋亡,抑制肿瘤增殖,延长荷瘤鼠的生存期。磁性纳米铁热疗能促进bax蛋白的表达,对bcl-2蛋白表达影响不明显。
Objective: To explore the efficacy of magnetic nano-iron hyperthermia on rat glioma.Methods: C6 cells were injected in the SD rat caudate nucleus by means of subculture of stereotactic inoculation.After 14 days when the glioma had growed to 0.5~1.0 cm in diameter,the rats were divided into four groups randomly and then the magnetic nano-iron(3,5,8 mg) or an equal volume of physic-ological saline were injected respectively.After finishing the magnetic nano-iron treatment for three times in alternating magnetic field and keeping 30 min for each time,the survival time of rats and the change of tumor volume were observed and the protein expression of bcl-2 and bax were detected by the method of immunohistochemistry SP.Results: The survival time of rats in the magheticnano-iron 3,5,8 mg groups was(19.60±1.43) days,(18.90±1.75) days,(20.40±2.26) days,respectivety.Which were significantly higher than that of control group[(13.30±1.69) days,P〈0.05].The expression of bax protein in the magheticnano-iron groups was also significantly stronger than that of control group(all P〈0.05).The expression of bax protein,bcl-2 protein and the survival time among the treatment groups had not difference(all P〉0.05).Conclusions: Magnetic nano-iron hyperthermia can effecttively promote cell apoptosis in rat glioma,inhibit tumor proliferation and prolong the survival time of tumor bearing rat.Magnetic nano-iron hyperthermia can promote the expression of bax protein,but the bcl-2 expression of protein is not obvious.
