目的探讨硒蛋白S(selenprotein S,SELS)单核苷酸多态性与鲍曼不动杆菌感染是否存在相关性。方法根据编码序列同义或非同义突变、频率〉5%的SNPs位点,以及文献报道的与细菌感染相关SNPs位点作为候选SNPs位点,采用多重高温连接酶检测反应(iMLDR技术)对解放军总医院第一附属医院和解放军总医院2010年1月-2012年12月收集的423例鲍曼不动杆菌感染病人和385例非感染病人的血标本进行SELS(rs4965373,rs4965814和rs34713741)SNP检测,对各基因位点基因型频数、等位基因频数及单倍型分布情况进行比较和分析。结果所有SNP符合哈迪-温伯格平衡。位点rs34713741的基因型与鲍曼不动杆菌感染有相关性(P〈0.05);位点rs4965373和rs4965814与鲍曼不动杆菌感染有相关性(P〉0.05),其单倍体型GT与鲍曼不动杆菌感染呈相关性(P=0.032)。结论硒蛋白S(SELS)SNPs位点(rs4965373,rs4965814和rs34713741)和鲍曼不动杆菌感染存在相关性。
Objective To determine the correlation of single nucleotide polymorphisms(SNP) of selenprotein S(SELS) and Acinetobacter baumanii infection in Chinese population. Methods Four hundred and twenty-three Acinetobacter baumanii infected patients and 385 patients collected from the same ward without infection were served as experimental group and control group, respectively. The candidate SNPs of SELS(rs4965373, rs4965814 and rs34713741) were selected according to their frequency(〉5%), as well as their associations with infectious diseases reported previously. SNP genotyping was performed using an improved multiplex ligation detection reaction(i MLDR) technique. The relation between Acinetobacter baumanii infection and SNPs of SELS were analyzed by multiple logistic regression model. Results All the 3 SNPs of SELS were in Hardy-Weinbeg equilibrium. The SNPs of rs34713741 was related to A. baumannii-infection(P 〈0.05), while the SNPs of rs4965373 and rs4965814 had no significant difference between the two groups(P 〉0.05), but the haplotype GT was identified to have significant association with Acinectobacter baumannii infection(P=0.032). Conclusion Our studies suggest that the SNPs(rs4965373, rs4965814 and rs34713741) are related to the susceptibility to Acinectobacter baumannii infection in a Chinese population.
