目的研究冬凌草甲素(oridonin)对高糖高脂诱导的人脐静脉内皮细胞(HUVECs)炎症反应的影响。方法体外培养HUVECs,将细胞分为空白对照组、高糖高脂组(给予33μmol/L葡萄糖+500μmol/L棕榈酸作用48h)、高糖高脂+2.5μmol/L oridonin预处理组(给予2.5μmol/Loridonin预处理后,给予高糖高脂处理,以下简称2.5μmol/Loridonin组)、高糖高脂+5μmol/L oridonin预处理组(给予5μmol/L oridonin预处理后,给予高糖高脂处理,以下简称5μmol/L oridonin组)。酶联免疫吸附法(ELISA)检测上清液中肿瘤坏死因子α(TNF—α)、白介素6(IL-6)的浓度,以观察细胞的炎症情况。结果高糖高脂组HUVECs细胞上清中炎症因子TNF—α、IL-6浓度明显高于空白对照组(t=14.77、14.05,P均〈0.01)。与高糖高脂组相比,2.5μmol/L oridonin组、5μmol/L oridonin组HUVECs细胞上清中炎症因子TNF-α(t=-4.81、-9.07,P均〈0.01)及IL-6(t=-4.23、-10.06,P均〈0.01)的浓度明显下降。结论冬凌草甲素能抑制高糖高脂诱导的HU—VECs细胞炎症反应。
Objective To investigate the effects of oridonin to inflammation factors of human umbilical vein endothelial cells (HUVECs) induced by high glucose and high fat. Methods The model of HUVECs was established and divided into 4 groups: control group, high glucose and high fat group(33 μmol/L glucose and 500 μmol/L,48 h) ,2. 5 μmol/L oridonin group and 5μmol/L oridonin group. The concentration of tumor necrosis factor - α (TNF - α )and interleukin -6 (IL- 6 )in the supernatant were detected by enzyme -linked immunosorbent assay (ELISA)to observe the inflammation state. Results The concentrations of TNF - α as well as IL - 6 in high glucose and high fat group were significantly higher than those in control group( t = 14.77,14.05 ,P 〈 0. 01 ). The concentrations of TNF - α( t = - 4.81, - 9.07 ,P 〈 0.01 ) as well as IL - 6 ( t = - 4.23, - 10.06, P 〈 0.01 ) in 2.5 μmoL/L oridonin group and 5 μmol/L oridonin group were significantly lower than those in high glucose and high fat group. Conclusion Oridonin can inhibit inflammation of HUVECs induced by high glucose and high fat.
