目的探讨肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)在卵圆细胞介导的肝再生中的作用。方法采用2-乙酰氨基芴(2-acetylaminofluroene,2-AAF)加2/3肝切除建立大鼠肝卵圆细胞介导的肝再生模型,以行2/3肝切除诱导的肝细胞介导的肝再生模型作为对照,采用RT-PCR方法检测TNF-α、IL-6在模型组及对照组肝组织中的表达。结果同正常组相比,术后模型组及对照组TNF-α mRNA表达明显上调,模型组持续至术后第3天(P〈0.05),对照组持续至术后第6天(P〈0.05)。对照组IL-6 mRNA术后8h显著升高(P〈0.05),而模型组IL-6mRNA术后无明显变化。结论TNF-α的高表达可能与肝卵圆细胞介导的肝再生过程密切相关。
Objective To investigate the role of TNF—α and IL-6 in oval celFmediated liver regeneration. Methods The model of oval cell-mediated liver regeneration was constructed by administration of 2-Acetylaminofluroene (2-AAF) combined with a two-thirds partial hepatectomy, and the regenerating liver tissues after simple 2/3 hepatectomy were taken as control. The mRNA of TNF—α and IL-6 in model group and control group was detected by RT-PCR. Results The levels of TNF—α mRNA in both model group and control group were significantly increased after hepatectomy. The level of TNF—α mRNA in model group was increased for only 3 days, while the level of TNF—α mRNA in control group increased for 6 days. The level of IL-6 mRNA in model group was significantly increased 8 h after hepatectomy, but the level of IL-6 mRNA in control group did not change significantly. Conclusion The higher expression of cytokine TNF—α might be significantly correlated with the hepatic oval cell-mediated liver regeneration.
