目的:优选山茱萸的水提醇沉多糖提取工艺条件。方法:以山茱萸多糖含量为指标,蒽酮比色法测定指标成分含量,在单因素试验基础上,采用L9(3^4)正交试验考察料液比、超声提取时间、重复提取次数和乙醇浓度对山茱萸多糖含量的影响,优化其提取工艺。并用DPPH自由基清除率法和总抗氧化实验对其抗氧化活性进行测定。结果:山茱萸多糖最佳提取的工艺条件为料液比1∶10,提取时间20 min,重复提取次数1次,提取乙醇浓度60%。实验显示,山茱萸多糖具有浓度相关的自由基清除能力和抗氧化能力。结论:优选的水提醇沉提取工艺稳定可行,所提取山茱萸多糖具有较强的自由基清除效果,是一种极具开发潜力的优良天然抗氧化剂。
Objective: To optimize the technology conditions of water extraction- alcohol precipitation for Cornus officinalis polysaccharides. Methods: With the content of Cornus officinalis polysaccharides as index tested by Anthrone colorimetry,based on the single factor test,effects of solid- liquid ratios,extraction time,repeat times and ethanol concentration were investigated by L9( 34) orthogonal experiment. The antioxidant activities were determined by the total antioxidant activity DPPH free radical scavenging experiment. Results: The research indicated that the optimum condition for extraction of Cornus officinalis polysaccharides was solid- liquid ratio 1: 10,extraction time 20 min,repeated 1 time,ethanol dose 60%. The ABTS free radical scavenging experiments showed that Cornus officinalis polysaccharides demonstrated the dose- dependent free radical scavenging and antioxidant activates. Conclusion: This optimized technology was stable and feasible. The isolated Cornus officinalis polysaccharides demonstrated great free radical scavenging activities,and thus providing a potential natural antioxidant for further development.
