目的:探究解毒胶囊对乳腺增生大鼠乳腺组织的形态、fasl和fas蛋白表达、caapase3mRNA表达的影响。方法:选取40只12周龄SD雌性未孕大鼠随机分为4组(正常组、模型组、方药组、三苯氧胺组),每组10只,造模成功后,经药物治疗4周,用HE染色观察各组乳腺组织形态,用免疫组化技术检测各组大鼠乳腺组织fas和fasl蛋白表达情况,用QPCR技术检测各组大鼠caspase3mRNA表达情况。结果:HE染色结果显示:模型组与正常组相比,乳腺腺泡腔面积变大;方药组和三本氧胺组与模型组相比,乳腺腺泡腔面积有明显减小。免疫组化结果显示正常组fas呈强阳性表达,模型组呈阴性或弱阳性表达,方药组和三苯氧胺组呈强阳性表达。而fasl表达情况与fas表达情况相反,正常组fasl弱阳性表达,模型组呈强阳性表达,方药组和三苯氧氨胺组呈弱阳性表达。QPCR结果显示模型组caspase3与正常组相比呈低表达(P〈0.1),治疗组、三本氧胺组与模型组相比呈高表达(P〈0.01),方药组和三苯氧胺组比起来没有统计学意义(P〉0.05)。结论:解毒胶囊能够干预乳腺增生大鼠的乳腺组织形态、乳腺组织的fasl fas蛋白表达、乳腺组织caspase3mRNA表达,对乳腺增生病有治疗作用。
Objective: To explore effects of Detoxification Capsule on morphology,fasl and fas protein expression,caspase3 mRNA expression of experimental hyperplasia rats' breast gland. Methods: We chose 40 female,12 weeks aged and non-pregnant SD rats and then randomly divided them into four groups: normal group,model group,herb group,tamexifen group. Each group had 10 rats. After the success of disease model,we treated them by drug. Use HE staining to observe each group' breast gland morphology. Use the technology about immunohistochemistry to test fasl,fas protein expression of each group's breast gland.Use QPCR technology to test caspase3 mRNA expression of each group's breast gland. HE staining. Results: the area of mammary gland acinus cavity in model group was bigger than that in normal group; the area of mammary gland acinus cavity in herb and tamoxifen group was smaller than that in model group. The expression of Fas in normal group was strong positive; the expression of fas in model was weak positive or negative; the result of fas in herb group and tamoxifen group was positive. But the result of fasl'expression was opposite to fas. The expression of fasl in normal group was weak positive or negative. The result of fasl expression in model group was strong positive. The result of fasl in herb and tamoxifen group was weak positive. The QPCR results showed that compared with the normal group,caspase3 expression of the model group showed lower expression( P〈0. 01). Compared with the model group,caspase3 expression of the rats' breast glands in the herbs group and the tamoxifen group showed higher and the herbs group's showed no statistical significance compared with the tamoxifen group' s( P〉0. 05). Conclusion: Detoxification Capsule can affect breast glands' morphology and intervene the expressions of fas,fasl and caspase3 in hyperplasia rats' breast glands. It has therapeutic effect on hyperplastic disease of breast.
