应用液相色谱串联质谱法(LC—MS—MS)鉴定富硒植物中四种有机含硒化合物,验证该方法的应用价值,了解这些富硒植物是否含有新的硒种类。植物样品用沸水提取,以甲醇-0.1%七氟丁酸水溶液(30:70)为流动相,滤液经Waters Symmetry C18(50mm×3.9mm,5μm)分离,经在线电喷雾离子源(ESI)正离子化后,采用多反应离子监测方式(MRM)测定含硒化合物。结果在4种富硒植物中检测到一到两种与标准品相同的含硒化合物,其它5种富硒植物中没有发现与标准品相同的有机硒。证明LC/MS/MS能有效、快速、准确筛查已有标准品的植物含硒化合物,在富硒植物中有望进一步获得新型含硒化合物。
To understand the practical application of LC-MS-MS method previously established for the analysis of γ-Glutamyl-Se-methylselenocyteine (GSeMC), methylselenocysteine (SeMC), selenocystine (Se-Cys), selenomethionine (SeMet) and the new selenium (Se) species in Se-enriched plants. Plant samples were extracted with deionized water in 100℃ and filtered. The filtrates were separated on Waters Symmetry C18(150 mm×3.9 mm, 5 μm)column with methanol-0.1% heptafluorobutyric acid (30:70) as mobile phase, at a flow rate of 500μl·min^-1. Electrospray ionization source(ESI) was applied and operated in positive ion mode. Multiple reactions monitoring (MRM) mode was used with nitrogen as collision gas. In nine samples, SeMC and/or SeMet were detected in four plants, five more plants did not contain the four selenocompounds. It is conclused that the method is quick, effective, specific and practical in the analysis of Se species with selenocompound standards in Se-enriched plants. New Se species are expected to be found in Se-enriched plants.
