目的:探讨养阴润目丸对干眼大鼠结膜上皮细胞肿瘤坏死因子-α(TNF-α)、核转录因子-κB(NF-κB)表达的影响。方法:SPF级雄性SD大鼠60只随机分成正常组、假手术组、养阴润目丸组、新泪然组和模型组,每组12只(24眼)。造模成功后,各组按相应处理方式连续干预3个月,实验结束后采用原位杂交法检测干眼大鼠结膜上皮细胞中TNF-αm RNA和NF-κB m RNA的表达。结果:连续干预3个月后,养阴润目丸组、新泪然组、模型组的SIT及BUT测定结果较正常组、假手术组为低(P〈0.01),其中,养阴润目丸组、新泪然组结果高于模型组(P〈0.01)。养阴润目丸组、新泪然组、模型组的结膜上皮细胞TNF-αm RNA和NF-κB m RNA的表达显著高于正常组、假手术组(P〈0.01),其中,模型组较养阴润目丸组、新泪然组表达更高(P〈0.01)。结论:养阴润目丸能够有效改善眼表症状,增加泪液分泌量,延长泪膜破裂时间,下调干眼大鼠结膜上皮细胞中TNF-α和NF-κB基因表达,这可能是其治疗干眼的重要机制之一。
Objective:To elucidate the effect of Yangyin Runmu Pill on expression of tumor necrosis factor-a(TNF-α)and nuclear factor kappa B(NF-κB) in conjunctival epithelium cells of rats with ophthalmoxerosis.Methods:Sixty male rats were randomly divided into normal control group,sham operation group,Yangyin Runmu Pill group,Tears Naturale Forte group and model control group,each group marked with A,B,C,D and E,respectively and containing 12 rats(24 eyes).Each group was given corresponding treatment for 3 consecutive months after modeling.The expression of TNF-α and NF-κB in conjunctival epithelium cells of rats with ophthalmoxerosis were detected by in situ hybridization.Results:After treatment for 3 months,the SIT and BUT in C,D,E groups were lower than those in A and B groups,with statistical significances(P〈0.01),of which,group C and D were higher than those in group E,with statistical significance(P〈0.01).The expression of TNF-α mRNA and NF-κB mRNA in conjunctival epithelium cells of C,D,E groups were higher than those in A and B groups,with statistical significances(P〈0.01) and group E were higher than those in C and D groups(P〈0.01).Conclusion:Yangyin Runmu Pill could improve eye symptoms,increase the tear secretion,prolong breakup time of tear film and down-regulate the expression of TNF-α and NF-κB in conjunctival epithelium cells of model rats with ophthalmoxerosis,and this may be one of the important mechanism for treatment of ophthalmoxerosis.
