目的:观察芪明颗粒对糖尿病肾病大鼠p-PKC、TNF-α表达的影响,以揭示芪明颗粒治疗糖尿病肾病的部分作用机理。方法:SD大鼠随机分为空白对照组(A组),模型对照组(B组),贝那普利西药对照组(C1组),黄葵中药对照组(C2组),芪明颗粒治疗组(D组),每组10只。于实验第8周末检测各组大鼠空腹血糖(FBG)、肌酐(Scr)、尿素氮(BUN),24 h尿微量白蛋白;采用光镜观察肾皮质形态学改变;采用ELISA法检测肾组织中TNF-α、p-PKC的水平。结果:(1)C1、C2、D组FBG、Scr、BUN、UAER均高于A组(P〈0.01),但明显低于B组(P〈0.01或P〈0.05)。(2)C1、C2、D组TNF-α、p-PKC水平较B组明显改善(P〈0.01)。(3)光镜下,C1、C2、D组肾脏病理改变较B组明显改善。结论:芪明颗粒能通过减少糖尿病肾病大鼠TNF-α、p-PKC水平减少尿白蛋白,改善肾功能和肾组织结构,对糖尿病大鼠肾脏有保护作用。
Objective : To observe the influence of Qiming particles on TNF - α and p - PKC expressions of diabetic nephropa- thy rats. Methods : SD rats were randomly divided into blank control group ( group A ), model control group ( group B), the western medicine of the control group (C1 group) ,Huangkui treatment group (C2 group) ,Qiming particles treatment group (D group), each group had 10 only. At the end of the 8th treatment week, FBG,Scr,BUN ,24 h urine trace albumin excretion rate (UAER) were tested, and then the cortex morphological change was observed. By ELISA test,TNF - oL and p - PKC levels in kidney tissues were measured. Results : ( 1 ) FBG, Scr, BUN and UAER in C1, C2 and D groups were higher than those in group A (P 〈 0. 01 ) , but were significantly lower than those in B group (P 〈0. 01 ). (2)TNF - α and p - PKC levels in C1 ,C2 and D groups were ob-viously improved than those in group B (P 〈0. 01 ). (3) By light microscopy,renal pathology changes in C1 ,C2 and D groups were obviously improved than that in group B. Conclusion : Qiming particles can reduce TNF - α and p - PKC levels of diabetic nephropathy rats and reduce urinary albumin, improve renal function and kidney organization structures, and protect diabetic rats kidneys.
