基于混合模式色谱-串联质谱联用技术,建立了一种简单、快速、灵敏的植物组织中内源性细胞分裂素(CKs)的分析检测新方法。采用大体积进样技术,提高了方法灵敏度。同时,由于混合模式色谱对CKs具有良好的分离能力,简化了前处理步骤。植物提取液经过C18固相萃取(SPE)小柱进行除杂,收集流出液并用氮气吹干后,复溶于700μL水中,进样540μL于高效液相色谱-串联质谱(HPLC-MS/MS)进行分析检测。该方法在1∽500pg/mL范围内线性关系良好,线性相关系数(r)大于0.9956;7种细胞分裂素的检出限(以3倍信噪比计算)在0.05∽0.23pg/mL之间;日内、日间相对标准偏差小于15.7%。所建立的方法已成功运用于不同植物组织和番茄幼苗中不同组织部位的内源性CKs种类及含量的分析。
A simple,rapid and sensitive method was developed for the determination of endogenous cytokinins(CKs)in plant samples using mixed-mode liquid chromatography-tandem mass spectrometry.By employing large volume injection technology,the sensitivity of detection was improved.As good separation of CKs was obtained with mixed-mode liquid chromatography,the sample pretreatment was simplified that its efficiency was improved.The crude plant extract was directly passed through a C18 SPE to remove impurity.Subsequently,eluent fraction was collected and evaporated to dryness under nitrogen stream.The residues were redissolved in 700μL water and 540μL was used for the analysis of CKs by HPLC-MS/MS.Good linearities were obtained for all 7CKs ranging from 1to 500pg/mL with correlation coefficients(r)higher than 0.9956.The LODs(Signal/Noise=3)for the CKs were found to be between 0.05 and 0.23pg/mL.Reproducibility of the method was evaluated by intra-day and interday measurements and the results showed that relative standard deviations(RSDs)were less than15.7%.Finally,the method was successfully applied for the determination of endogenous CKs in different plant tissues and different parts of Lycopersicum esculentum Mill.
