目的:观察电针抗高脂饮食诱导肥胖小鼠的作用及对脂肪组织炎症相关因子表达的影响。方法:选用40只刚断乳的SPF级C57BL/6J雄性小鼠,随机挑选8只普通饲料喂养为正常组,另一组32只高脂饲料喂养,将造模成功的16只随机分为模型组和电针组。比较各组小鼠体重、体脂、血脂等相关指标的差异,用HE染色观察比较各组小鼠脂肪的形态学差异,用实时荧光定量PCR检测各组小鼠脂肪组织中IL-6、TNF-α与MCP-1表达变化。结果:与模型组相比,电针组小鼠体重、内脏脂肪量下降明显;胆固醇、低密度脂蛋白含量均显著下降;脂肪组织IL-6、TNF-α、MCP-1的mRNA表达水平亦显著下降。结论:电针可以降低肥胖小鼠的体重、体脂,血脂,抑制肥胖小鼠脂肪组织炎症因子IL-6、TNF-α、MCP-1 mRNA的表达,改善肥胖小鼠炎症反应状态,从而实现减肥疗效。
Objective :To observe the effects of electro -acupuncture (EA) on weight reduction and the inflammatory factor in adipose tissues of the obese mice. Methods : Choose 40 SPF C57BL/6J male mice, and then randomly divide them into normal group (n=8) and high - fat group ( n = 32). After successful modeling, the obese mice ( n = 16) were randomly divided into model group (n =8) and EA group (n =8). Compare the differences in mice weight,fat,serum lipid of each group,the fat mor- phological differences among groups with HE staining. Use real -time PCR to detect the expressions of adipose IL -6 ,TNF -c-, MCP - 1. Results :The EA group's mice significantly decreased body weight, the visceral fat mass compared with those of the model group. The serum lipid, the level of cholesterol,low - density lipoprotein in the EA group decreased significantly compared with those of the model group. The expressions of IL - 6, TNF - α and MCP - 1 - mRNA in adipose tissue of EA group were signifi- cantly decreased compared with those of the model group. Conclusion:EA can reduce weight,visceral fat mass and serum lipid levels of the obese mice and inhibit the inflammatory factor in adipose tissue of obese mice,such as IL -6 ,TNF - α and MCP - 1. The effects of EA on fat reduction might be related to the inflammatory factor.
