建立了采用新型荧光试剂1,3,5,7-四甲基-8-苯基-(4'-O-(N-琥珀酰亚胺乙酸酯))-二氟化硼-二吡咯甲烷(TMPAB-OSu)为衍生化试剂的柱前衍生高效液相色谱分离荧光检测分析方法测定三种磷酸化氨基酸.在最佳的实验条件下,方法测定磷酸化丝氨酸(P-Ser)、磷酸化酪氨酸(P-Tyr)、磷酸化苏氨酸(P-Thr)的线性范围为0.05~2.5×10-6mol/L;检出限为2×10-9mol/L.
A sensitive detection method based on the derivatization of phosphoamino acids(P-Ser,P-Thr and P-Tyr) with 8-phenyl-(4-oxy-acetic acid N-hydroxysuccinimide ester)-4,4-difluoro-1,3,5,7-tetramethyl-4-bora-3a,4a-diaza-s-indacene(TMPAB-OSu) as the novel pre-column derivatization reagent followed by high performance liquid chromatography with fluorescence detection has been developed.Under the optimal conditions,the linear ranges are 0.05~2.5×10-6mol/L for P-Ser,P-Thr and P-Tyr respectively.The detection limits are 2×10-9mol/L respectively.
