脊髓神经元微小RNA-132在小鼠骨癌痛中的作用

目的评价脊髓神经元微小RNA-132(miR-132)在小鼠骨癌痛中的作用。方法雄性C3H/HeJ小鼠,4~6周龄,体重20~25g。第一部分实验C3H/HeJ雄性小鼠52只,采用随机数字表法,将其分为2组(n=26):假手术组(S组)和骨癌痛组(BCP组);第二部分实验C3H/HeJ雄性小鼠32只,采用随机数字表法,将其分为4组(n=8):假手术+鞘内注射无核酸酶水组(S+Veh组)、骨癌痛+鞘内注射无核酸酶水组(BCP+Veh组)、骨癌痛+鞘内注射阴性对照锁核酸组(BCP+c组)和骨癌痛+鞘内注射miR-132反义锁核酸组(BCP+AS组)。采用右侧股骨远端骨髓腔注射2×10^5个NCTC2472细胞的方法制备骨癌痛模型。BCP+c组及BCP+AS组于接种后14d,分别鞘内注射阴性对照锁核酸和miR-132反义锁核酸12pmol/5μl,S+Veh组及BCP+Veh组给予等容量无核酸酶水,连续7d,1次/d。S组和BCP组分别于接种前、接种后4、7、10、14、21d时测定机械缩足反应阈(MWT)和自发抬足次数(NSF),并测定脊髓水平miR-132表达;S+Veh组、BCP+Veh组、BCP+C组和BCP+AS组于接种后14d鞘内给药前、接种后15、16、17、18、19、20、21d时测定上述痛行为学指标。结果与s组比较,BCP组接种后7-21d时MWT降低,NSF增加,脊髓水平miR-132表达上调(P〈0.05);与s+Veh组比较,其他3组各时点MWT降低,NSF增加(P〈0.05);与BCP+Veh组比较,BCP+AS组接种后16.21d时MWT升高,NSF减少(P〈0.05)。结论脊髓神经元miR-132参与了小鼠骨癌痛的形成和维持。

英文摘要:

Objective To evaluate the role of spinal neuronal microRNA-132 (miR-132) in a mouse model of bone cancer pain (BCP). Methods Male C3H/HeJ mice, aged 4-6 weeks, weighing 20-25 g, were used in the study. The experiment was performed in 2 parts. In experimentⅠ, fifty-two male C3H/HeJ mice were randomly divided into 2 groups (n = 26 each) using a random number table: sham operation group (S group) and BCP group. In experiment Ⅱ , thirty-two male C3H/HeJ mice were randomly divided into 4 groups ( n = 8 each) using a random number table: sham operation + intratheeal DNAse/RNAse-free water group (S + Veh group), BCP + intratheeal DNAse/RNAse-free water group (BCP + Veh group), BCP + intrathecal negative control locked nucleic acid (LN~) group (BCP + C group) , and BCP + intratheeal miR-132 antisense LNA group (BCP + AS group). BCP was induced by injecting 2 ×10^5 NCTC 2472 cells into the distal medullary cavity of the right tibia. In BCP + C and BCP + AS groups, negative control LNA and miR-132 antisense LNA 12 pmol/5 μ1 were intrathecally injected, respectively, once a day for 7 consecutive days starting from day 14 after inoculation. In S + Veh and BCP + Veh groups, DNAse/RNAse-free water 5 μl was intrathecally injected once a day for 7 consecutive days starting from day 14 after inoculation. In S and BCP groups, the paw withdrawal threshold to mechanical stimuli (MWT) and the number of spontaneous flinches ( NSF) were measured, and the expression ot miR-132 in spinal cord was detected before inoculation and on 4, 7, 10, 14 and 21 days after inoculation. In S + Veh i BCP + Veh, BCP + C and BCP + AS groups, the parameters mentioned above were measured at 14 days before inoculation and 15, 16, 17, 18, 19, 20 and 21 days after inoculation. Results Compared with S group, MWT was significantly decreased, NSF was increased and the expression of miR-132 was up-regulated on 7-21 days after inoculation in BCP group. Compared with S + V
