目的:探讨电针大鼠胃经穴对胃黏膜细胞细胞外信号调节激酶(ERK)免疫组化的影响。方法:72只大鼠随机分为6组,利用链霉蛋白酶消化法分离胃黏膜细胞,分别用PD153035和100mL/L血清孵育胃黏膜细胞,免疫组化法检测ERK的表达。结果:胃经组和胆经组胃黏膜细胞ERK的表达明显高于正常组和模型组(P〈0.01);胃经组胃黏膜细胞ERK的表达与胆经组比较显著增强(P〈0.01);胃经+PD153035组胃黏膜细胞ERK的表达明显低于胃经组(P〈0.01)。结论:电针能上调胃黏膜细胞ERK的表达,ERK可能是胃经与胃相关的客观物质基础之一。
Objective:To observe the expression of the Extracelluar regulated protein kinase(ERK) immunity histochemistry on gastric mucosal cells in rats with electro-acupuncture at stomach meridian acupoints.Methods:72 rats were randomly divided into six groups,Water-immersion and restrained stress methods was adopted for stress-induced gastric mucosal injury rat model,gastric mucosal cells were separated by using pronase digestion method,and incubated by PD153035,a EGFR inhibitor and 100ml/L serum,ERK expression in gastric mucosal cells was tested by Immunity Histochemistry.Results:In comparison with those in the Normal group or Model group,the ERK expression in gastric mucosal cell in the Stomach meridian group and Gallbladder meridian group were significant reinforcement(P0.01).Furthermore,the ERK expression in Stomach meridian group were more obvious than those in Gallbladder meridian group(P0.01).Compared with Stomach meridian group,the reduction of the ERK expression in Stomach plus PD153035 group was more significant(P0.01).Conclusions:The electro-acupuncture can activate the ERK expression in gastric mucosal cells,ERK is one of the key material of the relative particularity between Stomach Meridian and Stomach.
