副猪嗜血杆菌病(Haemophilus parasuis)是规模化猪场的重要细菌性疾病之一。对已发表的HPS外膜蛋白P5序列进行相关序列分析,设计合成引物,以HPS血清5型基因组为模板,通过PCR扩增HPS外膜蛋白P5编码基因,获得1 116 bp的预期基因片段。该基因编码372个氨基酸的蛋白质,预测分子量大约为41 kDa,利用基因工程的方法将该基因片段定向克隆至表达载体pGEX-6p-1中,诱导表达后的分子量约为67 kDa。该研究为制备亚单位疫苗和诊断试剂奠定了基础。
Haemophilus parasuis disease is one of an important bacterial diseases in large-scale pig farm.Analysis the sequence of published outer membrane protein P5,designed and synthesized the primers,took the HPS serotype 5 genome as a template,amplified the outer membrane protein P5 gene of HPS by PCR,and got the expected 1 116 bp fragment,the gene encoding 372 amino acids,the Prediction molecular weight of it about 41 kDa,We used the genetic engineering technology cloned gene into the expression vector pGEX-6p-1,and the induced molecular weight of ompP5 about 67 kDa.The study prepared a basis of subunit vaccine and diagnostic reagents.
