胡萝卜是一种具有重要保健功能的蔬菜,其块状根系富含胡萝卜素等多种营养物质,可开发作为生物反应器。以转基因胡萝卜根作为生物反应器生产生物活性成分需要应用其根部高效、特异性表达的启动子。本研究应用cDNA-AFLP分析胡萝卜根、叶中基因的差异表达,获得32个在根中高效表达而在叶中不表达的转录衍生片段(TDF)。测序和同源比对分析结果表明,其中有20个TDF在基因库中未发现同源基因,其他12个分别是推定的植物激素抑制蛋白、金属硫因-1蛋白及其他有关基因。
Daucus carota is one of the most important vegetables with high healthy value, its root has various nutrient substances including plenty carotin, so it can be developed to biological reactor. To produce material with biological activity by biological reactor of transgenic D. carota root, specific and high efficiency promoters are needed. In this paper, gene variant expression in root, stem and leaf of D. carota is analyzed by cDNA-AFLP, 32 TDFs highly-efficient expressed in root and non-expressed in stem and leaf were found. The results of DNA sequences and homologous alignment showed that homologous gene was not found in 20 TDFs, the other 12 TDFs are turned out to be inhibitory protein of plant hormones, metallothionein-1 protein and other relative genes.
