AIM ：To study the effect of IL - 10 on the expression of monocyte chemotactic protein - 1 （ MCP -1 ） in human dental follicle cells （HDFCs）. METHODS： HDFCs of the 5th passage were co - cultured with 25ng/mL IL - 10 for 0, 1, 3, 6, 9 and 12 h. The mRNA expression of MCP - 1 was detected by RT - PCR. In the next experiment the cells were co - cultured with 25ng/mL IL - 10 for 0, 2, 4, 6 and 8h. The protein expression of MCP - 1 was examined by ELISA. RESULTS ： 25ng/mL IL - 10 down - regulated the mRNA expression of MCP - 1 in 1 - 6h（ P 〈 0.05 ） and the protein expression of MCP - 1 in 4 - 8h （ P 〈 0.05 ）. CONCLUSION： IL - 10 inhibits MCP - 1 expression in HDFCs. Since MCP - 1 is an important factor to recruit mononuclear cells, IL - 10 suppresses the recruitment of these cells to the follicle by down - regulating MCP - 1 expression.
