目的通过研究丹皮酚对TNF-α诱导的真皮成纤维细胞MMP-9mRNA及细胞因子表达的影响,探讨丹皮酚抗皮肤炎症的可能机制。方法采用RT—PCR法考察正常人真皮成纤维细胞中MMP-9mRNA表达情况以及丹皮酚对TNF-α诱导的真皮成纤维细胞MMP-9mRNA表达的影响。采用ELISA法测定丹皮酚对TNF—α诱导的真皮成纤维细胞表达IL-1β、IL-6及IL-8的影响。结果MMP-9在正常真皮成纤维细胞上表达极弱,TNF-α可以诱导MMP-9mRNA的表达,丹皮酚可以抑制TNF—α诱导引起的MMP-9mRNA表达的上调。成纤维细胞可分泌少量的IL-1β、IL-6及IL-8,TNF—α可以明显增加成纤维细胞IL-1β和IL-8的分泌量,丹皮酚可以抑制TNF—α诱导的IL-1β和IL-8的产生,但对IL-6的分泌无影响。结论丹皮酚可以抑制TNF—α诱导真皮成纤维细胞引起的MMP-9mRNA及炎症因子IL-1β和IL-8表达水平的上调。
Aim To explore the possible anti-inflammation mechanism of paeonol by investigating its effects on the MMP-9 mRNA expression and cytokines production in human dermal fibroblasts induced by TNF-α. Methods The effect of paneonol on the expression of MMP-9 mRNA was detected by semi-quantitative RT- PCR. The modulation of paneonol on the production of IL-1β, IL-6 and IL-8 in fibroblasts was measured by ELISA. Results MMP-9 hardly expressed in human dermal fibroblast. The results also showed that TNF-α significantly induced the expression of MMP-9 in fibroblasts and at the same time paeonol inhibited the expression of MMP-9. TNF-α stimulated the production of IL-1β and IL-8 in fibroblasts, while 10 -100 mg · L^-1 paeonol inhibited TNF-α-induced IL-1β and IL-8 production in fibroblasts but had nothing to do with the production of IL-6. Conclusions Paeonol can inhibit the expression of MMP-9 mRNA, IL-1β and IL-8 induced by TNF-α.
