目的检测白细胞介素-6(IL-6)在人脑胶质瘤干细胞(GSCs)中的表达及其对GSCs侵袭力的影响。方法从8例人脑胶质瘤中培养出GSCs并予以鉴定。实时定量RT—PCR及酶联免疫吸附试验法检测IL-6在GSCs以及相应原代培养胶质瘤细胞(PGCs)中的表达水平并比较分析;使用中和性IL-6抗体或外源性IL-6后进行基质胶侵袭实验,检测GSCs及相应PGCs侵袭力的变化并比较分析。结果配对t检验结果显示IL-6在GSCs中mRNA和蛋白表达水平均显著高于相应的PGCs(分别为t=2.578,v=7,P〈0.05和t=4.690,v=7,P〈0.005);基质胶侵袭实验中,加入IL-6抗体可以明显减低PGCs和GSCs的侵袭力(分别为t=3.105,v=7,P〈0.05和t=13.922,v=7,P〈0.001);而外源性IL-6则可增加PGCs和GSCs的侵袭力(分别为t=-5.983,v=7,P〈0.001和t=-6.935,v=7,P〈0.001)。结论IL-6在GSCs中高表达,并且在体外实验中与PGCs和GSCs的侵袭性相关。IL-6可能在胶质瘤免疫抑制及肿瘤侵袭中起关键作用。
Objective To investigate the expression of interleukin-6 in glioma stem cells (GSCs) and its role in glioma stem cells invasion. Method GSCs from eight surgical glioma specimens were cultured and identified. Real-time RT-PCR and Enzyme-Linked Immunosorbent Assay were used to measure and compare the expression levels of IL-6 in GSCs and corresponding primary glioma cells (PGCs). Subsequently, IL-6 antibody or exogenous IL-6 was used in a Matrigel-invasion assay and effects of IL-6 on GSCs or PGCs invasiveness were then determined. Results Paired t-test showed that IL-6 mRNA and protein expression levels were significantly higher in GSCs than in PGCs from the identical tumor ( t =2.578,v =7, P 〈0.05 and t =4.690, t, =7, P 〈 0. 005, respectively). In Matrigel-invasion assay, PGCs and GSCs invasiveness markedly decreased after IL-6 was blocked with neutralizing antibody (t =3.105, v =7, P 〈0.05 and t = 13. 922, v =7, P 〈0.001, respectively), but significantly increased when exogenous IL-6 was added ( t = -5. 983, v =7, P 〈0. 001 and t = -6. 935, v = 7, P 〈 0. 001, respectively). Conclusions The present study found that the IL-6 is overexpressed in GSCs and significantly augments GSCs invasiveness in vitro, suggesting a potential target in future therapy for GSCs rather than for their derivatives.
