目的探讨外源性Neogenin对人滋养细胞凋亡的影响及其凋亡机制中可能存在的传导通路。方法行早孕期滋养细胞株TEV-1培养,应用吖啶橙/溴化乙啶(AO/EB)双染法检测经不同浓度的Neogenin处理细胞24 h后TEV-1细胞的凋亡情况,并同时检测Caspase-3活性的变化。结果 Neogenin可诱导TEV-1细胞凋亡,且存在剂量依赖性。在0、1、5、10及50 ng/mL浓度Neogenin作用下,其凋亡率依次为(5.34±0.32)%、(29.86±0.92)%、(38.15±0.24)%、(40.15±0.27)%及(52.37±0.20)%。但Caspase-3的活性变化不明显,依次为对照组的(1.02±0.02)、(1.06±0.00)(、0.93±0.02)(、1.04±0.01)倍。结论 Neogenin诱导滋养细胞凋亡,其凋亡信号通路与Caspase信号传导通路不存在必然联系。
Objective To study the effect of Neogenin on apoptosis of trophoblast cells and the possible pathway in the mechanism of apoptosis.Methods TEV-1 cells in exponential growth stage were treated by different doses of Neogenin for 24 h.Apoptosis of TEV-1 cells was assessed by AO/EB assay.The relative Caspase-3 activity was measured by colorimetric assay.Results Neogenin could induce apoptosis of TEV-1 cells in a dose-dependent way.Under the action of different concentrations of Neogenin with 0,1,5,10 and 50 ng/mL,the apoptosis rate was(5.34±0.32)%,(29.86±0.92)%,(38.15±0.24)%,(40.15±0.27)% and(52.37±0.20)% respectively.The relative activity of Caspase-3 had no significant change and was(1.02±0.02),(1.06±0.00),(0.93±0.02) and(1.04±0.01) fold of that in control group.Conclusion Neogenin could induce apoptosis of TEV-1 cells without involving Caspase-transduced signal pathway.
