胆囊收缩素对砷中毒大鼠热刺激缩足反射潜伏期的影响

目的:探讨皮下注射八肽胆囊收缩素(CCK-8S)对砷中毒大鼠热刺激缩足反射潜伏期(paw withdraw ther-mal latency,PWTL)的影响。方法:wistar雌鼠32只,体重180-220g,出生8-10周,随机分为四组,A组(n=8):尾静脉注射生理盐水组;B组(n=8):尾静脉注射亚砷酸钠(NaAsO2)组;C组(n=8):尾静脉注射NaAsO2+皮下注射CCK-8S组;D组(n=8):尾静脉注射NaAsO2+皮下注射生理盐水组。在注射前、注射后2、4、6、8、10、12、14、16天分别进行热板实验,记录PWTL。在PWTL达到最小值时,C组皮下注射CCK-8S后,D组皮下注射生理盐水,分别在注射前及注射后15、30、45、60、759、0分钟进行热板实验。结果:在注射NaAsO2后第8天,B组的PWTL与A组及注射NaAsO2前相比较差异均具有显著性(P〈0.01),砷中毒模型制备成功。在第8天,发现C组PWTL在15-60分钟时与注射CCK-8S前及D组比较均显著的延长(P〈0.05)。结论:CCK-8S能显著降低砷中毒大鼠的热痛觉过敏,对砷中毒大鼠有镇痛作用。

英文摘要:

Objective: To investigate the effect of cholecystokinin octapeptide on PWTL of arsenic- poisoning rat. Methods:Thirty two female Wistar rats (8 - 10 weeks)weighing 180 - 220 g were randomly divided into four equal groups, each group 'has 8 animals: group A inject 0. 9 % sodium chloride at 1ml/100g, group B received NaAsO2 injection, group C inject NaAsO2 first, then CCK- 8S at 0.05mg/kg via subcutaneous,group D received NaAsO2 first, then 0. 9 % sodium chloride via subcutaneous. The NaAsO2 injection and 0. 9 % sodium chloride of A were injected via caudal vein. The PWTL was measured by a hot plate test (55℃)before NaAsO2 injection in 2,4,6 ,8 , 10 , 12 , 14,16 days . While the PWTL achieved minimum value, group C inject CCK - 8S and group D inject 0. 9 % sodium chloride via subcutaneous. The PWTL of group C and group D was measured bcfore and 'after injection in 15,30,45,60,75,90 minutes. Results:Before experiment there was no significant difference in PWTL between group A and group B(P 〉 0.05), after 8 days there is significant difference in PWTL between group A and group B(P〈 0.01 ), these data have show that subacute arsenic-poisoning rat model was successfully induced. After CCK- 8S injection there was significant prolong in PWTL in group C between 15 min and 60 min compared to before injection and group D ( P 〈 0.05). Conclusion: This study shows that CCK - 8S significant reduces the thermal hyperalgesia of arsenic-poisoning rat CCK - 8S have analgesic effect to arsenic-poisoning rat.
