目的:探寻清肠化湿方对小鼠溃疡性结肠炎急性活动期模型结肠组织黏膜修复及COX-2表达的影响,进而分析其可能的作用机制。方法:采用TMBS法制备小鼠溃疡性结肠炎急性活动期模型。实验设对照组、模型组、清肠化湿方低剂量组,清肠化湿方中剂量组,清肠化湿方高剂量组,造模前预防性给药3天,造模后给药3天,分别观察给药结束后小鼠结肠组织黏膜修复及COX-2表达情况。结果:清肠化湿方低、中、高剂量组与模型组比较,结肠组织炎症程度、黏膜糜烂、溃疡等都有所减轻,且各组病理学评分显著低于模型组,有显著性差异;模型组小鼠结肠组织COX-2表达显著增强,清肠化湿方各剂量组可明显降低COX-2表达,清肠化湿方低、中剂量组与模型组比较有显著性差异。结论:清肠化湿方能修复溃疡性结肠炎结肠黏膜,抑制溃疡性结肠炎小鼠结肠组织COX-2的表达,可能是其疗效的机制之一。
Objective:To study the prevention and treatment effect of Qichang Huashi Herbal Decoction repairing the colonic mncosa and influencing COX-2 expression on rat models of acute ulcerative colitis, and to explore the possible mechanisms of the Qichang Huashi Decoction's therapeutic effect. Methods :We used TNBS/anhydrous alcohol to make model. Then, model rats were divided into normal group, model group, the low, the middle and the high dose Qichang Huashi Decoction groups. Before modeling, Qichang Huashi Decoction was used for prevention for 3 days and after modeling to give drug dilutions for 3 days. Then, all groups of rats were killed and colon tissues were cut out for inspections. Results : Compared with the model group, the low, the middle and the high dose of Qichang Huashi Decoction groups, the degree of inflammation of the colon tissue, mucosal erosions and ulcers were reduced, and the pathological score in each group was significantly lower than that in the model group. In the model group,COX-2 expression of colon tissue was significantly enhanced,while the low, the middle dose of Qichang Huashi Decoction groups can significantly reduce the expression of COX-2. Conclusion:Qichang Huashi Decoction can repair /he colonic mueosa of ulcerative colitis and suppress the COX-2 expression in ulcerative colitis colon tissue. That is the one of the mechanisms to treat UC.
