三七总皂苷对顺铂诱导肾损伤大鼠模型自噬蛋白LC3和Beclin1的影响

目的研究三七总皂苷(PNS)对顺铂诱导肾损伤大鼠肾组织中自噬相关蛋白LC3、Beclinl表达的影响。方法将36只雄性SD大鼠随机分为空白对照组,顺铂模型组,顺铂+PNS组,在给药24h后处理动物,检测血清肌酐(Cr)、尿素氮(BUN)和β-N-乙酰胺基葡萄糖苷酶(NAG)水平,利用透射电镜观察大鼠肾小管上皮细胞粒体形态及自噬体,通过Westernblot检测自噬蛋白LC3和Beclinl蛋白的表达。结果大鼠暴露于顺铂24h后,与空白对照组比较,顺铂模型组血清Cr、BuN显著升高(P〈0.05),尿NAG水平没有明显变化,电镜观察到自噬体出现,肾组织LC3和Beclinl蛋白表达显著升高(P〈0.01)。与顺铂模型组比较,顺铂+PNS组血清cr明显降低(P〈0.05),LC3和Beclinl蛋白表达显著升高(P〈0.01)。结论PNS可能通过提高Beclinl的表达,增强肾组织的自噬而降低顺铂诱导的大鼠肾损害。

英文摘要:

OBJECTIVE To explore the effect of Panax notoginseng saponins (PNS) on the expression of autophagy-related protein LC3 and Beclinl in renal tissues of rats with cisplatin-induced renal damage. METHODS 36 male SD rats were randomly divided into the control group, model group and PNS group. 24 h after administration, the animals were sacrificed and the levels of serum creatinine (Cr), blood urea nitrogen (BUN) and urine β-N-aeetamido-glucosidase (NAG) were determined. The morphology of renal tubular ep- ithelial cells and mkochondrial autophagy were observed under transmission electron microscopy. The expression of autophagy protein LC3 and Beclinl protein were detected by Western blot. RESULIS Compared with the control group, the levels of serum Cr and BUN significantly increased in model group (P〈0.05); however, the level of urinary NAG did not change obviously. Autophagy was observed under electron microscopy and the expression of LC3 and Beclinl in kidney tissue significantly increased (P〈0.01). Corn pared with the model group, serum Cr was significantly reduced in PNS group (P〈0.05), but the expression of LC3 and Beclinl sig- nificantly increased (P〈0.01). CONCLUSION PNS can reduce cisplatin-induced kidney damage in rats by enhancing autophagy in renal tissue and increasing the expression of Beclinl.
