目的 探讨阿托伐他汀对放射性心脏损伤的保护作用.方法 选择SD大鼠45只,随机分为对照组、X线照射组、阿托伐他汀组,每组15只,其中X线照射组、阿托伐他汀组予以10 Gy/d X线照射,共3d.大鼠照射完毕处死后,测定血清肌钙蛋白Ⅰ(cTnⅠ)水平,心肌组织TNF-αmRNA及蛋白的表达.结果 与对照组比较,X线照射组和阿托伐他汀组血清cTnⅠ[(2.14±0.14)ng/ml,(1.85±0.12)ng/ml vs (0.59±0.06) ng/ml]、心肌组织TNF-αmRNA及蛋白表达明显升高(P<0.05);与X线照射组比较,阿托伐他汀组血清cTnⅠ、心肌组织TNF-α mRNA及蛋白表达明显降低(P<0.05).结论 阿托伐他汀能使心肌组织TNF-αmRNA及蛋白的表达下调,从而有效减轻大鼠放射后心脏炎性反应,减轻心脏损伤.
Objective To study the effect of atorvastatin on heart injury following irradiation.Methods Forty-five SD rats were randomly divided into control group,X-ray irradiation group,and atorvastatin treatment group (15 in each group).The animals in X-ray irradiation group and atorvastatin treatment group received X-ray irradiation (10 Gy/d) for 3 days and then killed.Serum troponin Ⅰ (cTnⅠ) level and TNF-α protein and mRNA expression level in their myocardial tissue were measured.Results The serum cTnⅠ level and TNF-α mRNA and protein expression level were significantly higher in X-ray irradiation group and atorvastatin treatment group than in control group (2.14±0.14 ng/ml,1.85±0.12 ng/ml vs 0.59±0.06 ng/ml) and significantly lower in atorvastatin treatment group than in X-ray irradiation group (P<0.05).Conclusion Atorvastatin can effectively reduce the inflammatory reaction and heart injury in rats following irradiation by down-regulating the expression of TNF-α mRNA and protein.
