目的探讨吡格列酮对大鼠冠状动脉微栓塞(CME)后心肌损伤的影响。方法经左心室注射42μm微栓塞球建立CME模型后存活的雄性sD大鼠30只,随机分为单纯微栓塞组(CME组)与吡格列酮预处理组(PIO组),每组15只。PIO组术前连续7d给予吡咯列酮10mg/(kg·d)灌胃。另选择经左心室注射等量生理盐水后存活的雄性SD大鼠15只为假手术组(S组)。术后12h应用RT-PCR及免疫印迹法检测各组大鼠心肌组织炎性因子及抗炎因子的表达。结果与S组比较,CME组和PIO组白细胞介素-6(IL-6)、白细胞介素-10(IL-10)的mRNA表达水平、肿瘤坏死因子-α(TNF-α)mRNA及蛋白表达水平明显升高(P〈0.05)。与CME组比较,PIO组IL-6mRNA、TNF-αmRNA及蛋白表达水平明显降低(P〈0.05)、IL-10mRNA表达水平明显升高(P〈0.05)。结论吡咯列酮预处理可明显改善CME后心肌炎性反府.其作用机制可能与下调炎性因子表达有关。
Objective To investigate the effect of Pioglitazone (PIO) on rats with myocardial injury after coronary mi- croembolization (CME), Methods 30 survival of male SD rats were randomly divided into pure CME group (CME group) and PIO pretreatment group (PIO group), and these rats were injected with mierosphcres (42 μm in diameter) into the left ventricles to induce the model of CME, each group of 15 rats. PIO group wasreceived PIO 10 mg/(kg.d) for 7 days before operation, Another 15 survival of SD rats were selected as sham operation group (S group), and these rats were injected with the same dose of normal saline by left ventricles. The expression on inflammatory cytokines and anti-in- flammatory factor of myocardial tissue of rats in each group were detected by RT-PCR and Western Blot after operation of 12 h. Results Compared with S group, IL-6 mRNA, IL-10 mRNA, TNF-α mRNA and TNF-α protein expression level of CME group and PIO group were significantly elevated (P 〈 0.05). Compared with CME group, IL-6 mRNA, TNF-α mRNA and TNF-α protein expression level of PIO group were significantly decreased, IL-10 mRNA expression level of PIO group was significantly elevated (P 〈 0.05). Conclusion PIO pretreatment can significantly improve CME- induced myocardial injury via downregulation of inflammatory cytokines.
