丹参酮ⅡA分子印迹聚合物的制备及其丹参活性成分的分离

目的制备丹参酮ⅡA的分子印迹聚合物(MIP)及分离丹参的活性成分。方法以丹参酮ⅡA为模板,以α-甲基丙烯酸(MAA)为功能单体,乙二醇二甲基丙烯酸酯(EDMA)为交联剂,采用热聚合方式,通过条件优化获得了对丹参酮ⅡA有特异选择性吸附的MIP。通过静态平衡结合法研究了模板聚合物的结合能力和选择特性,用Scatchard分析法研究了MIP对模板分子的结合特性。结果制备MIP最优条件为ⅡA-EMAA-EDMA(1:4:24),溶剂为氯仿。该聚合物形成了两类不同的结合位点,高亲和力结合点的离解常数Kd1为0.752 mmol·L^-1、最大表观结合常数Qmax1为21.58μmol·g^-1,低亲和力的Kd2为2.371 mmol·L^-1、Qmax2为44.63μmol·g^-1。结论该MIP可用于分离丹参酮提取物中的丹参酮ⅡA,效果良好。

英文摘要:

OBJECTIVE To preparation of Tanshinone ⅡA imprinted polymer and its application in separation of active ingredients in salvia extracts. METHODS Molecularly imprinted polymers (MIP) of Tanshinone Ⅱ A with recognition properties were prepared by thermal polymerization method using Tanshinone Ⅱ A as templates, α - methyacrylic acid as functional monomers and ethylene dimethacrylate as crosslinker under the optimized conditions. The binding experiments of MIP to Tanshinone ⅡA were used to evaluate the influences of various factors. Equilibrium binding experiment was used to investigate the binding ability to template molecule and the substrate selectivity. The scatchard analysis was used to study the binding characteristics of MIP to template molecule. RESULTS The optimized conditions were Tanshinone ⅡA -MAA -EDMA (1 : 4 : 24 )with chloroform as the solvent. The MIPs recognized template with two classes of binding sites. Scatchard analysis showed that the dissociation constant Kd1 and apparent maximum number Q of binding sites with high affinity are 0. 752 mmol·L^-1 and 21.58 μmol·g^-1 , while Kd2 and Qmax2 of the binding sites with low affinity are 2.371 mmol· L^ - 1 and 44.63μmol· g^- 1, respectively. CONCLUSION The imprinted polymer was applied to separate Tanshinone ⅡA from Tanshinone extracts and good results were obtained.
