利用毛细管电泳-安培法检测联用技术,建立了一种简单、灵敏、快速同时分离检测人体尿液中两种β-阻断剂类兴奋剂药物索他洛尔和塞利洛尔的新方法。以直径300μm的铂圆盘电极为检测电极(铂丝为辅助电极,Ag/AgCl为参比电极),在0.950V的检测电位下,30mmol/L Clark-Lubs缓冲体系(pH9.60)中,两种分析物在8min内达到基线分离。在最佳条件下,其峰电流与0.15~60mg/L索他洛尔与0.30~60mg/L塞利洛尔呈现良好的线性关系,检出限分别为0.05mg/L和0.09mg/L,并将建立的方法成功地应用于人体尿样的分析。进~步研究了人体尿液中索他洛尔36h内的排泄曲线及索他洛尔在尿液中的排泄规律,对奥运会中的兴奋剂快速检测具有一定的应用价值。
A simple, sensitive, and quick method was established to separate and determine two β-blockers, sotalol and celiprolol in human urine by capillary electrophoresis-amperometric detection. Under optimal conditions of the detection potential of 300 μm platinum disk electrode at O. 950 V (vs. Ag/AgCl), pH 9.60, 30 mmol/L Clark-Lubs buffer solution, the two analytes were baseline separated in 8 min. The calibration curves exhibited an excellent linear behavior over the concentration range about three orders of magnitude. The detection limits were 0.05 and 0.09 mg/L for sotalol and celiprolol, respectively. The method was successfully applied to the detection of sotalol in the human urine after 36 h of administration of sotalol tablets, and the metabolism curve of it was also obtained.
