采用毛细管区带电泳-安培检测法建立了同时测定中草药天仙子中主要成分阿托品和东茛菪碱的新方法.在电极电位为1.000V,毛细管运行液为50mmol/L磷酸盐缓冲溶液(PBS)(pH为8.0)中,当分离电压为12kV时,阿托品和东莨菪碱2个组分在8min内快速达到基线分离.阿托品和东莨菪碱的峰高和浓度在1.0—40mol/L(阿托品)和3.0~100mol/L(东莨菪碱)范围内呈良好的线性关系,检测限均为7.0×10^-7mol/L,并具有很好的重现性(RSD小于3.3%).
Atropine and scopolamine in Hyoscyamus niger by Capillary Electrophoresis - amperometric detection was studied. Under the following optimal conditions: detection potential of 1. 000 V ( vs Ag/ AgCl), 50 mmol/L phosphate buffer solution at pH 8.0 and separated voltage of 12 kV; the two analytes were baseline separated in 8 min. The linear ranges were 1.0 - 40 mol/L and 3.0 - 100 mol/L for atropine and scopolamine, respectively. And the detection limits of both components were 7.0 × 10^-7 mol/L. The RSD were less than 3.3%.
