目的:通过研究隐丹参酮对多囊卵巢综合征(PCOS)高雄激素血症的干预作用及可能的调控机制,为临床治疗PCOS高雄激素血症新药物的探索提供科学依据。方法:①体内实验研究隐丹参酮对PCOS高雄激素血症动物模型生殖内分泌功能的影响;②体外实验研究隐丹参酮对高雄激素血症细胞模型雄激素分泌功能的影响;③采用免疫组化及real.timePCR法对雄激素合成代谢相关酶及其编码基因进行检测。结果:隐丹参酮可以改善模型大鼠的动情周期、体质量、卵巢系数及卵巢形态,逆转大鼠血清睾酮(T),雌烯二酮(A2),促黄体生成素(LH),性结合球蛋白(SHBG)水平;抑制模型细胞株的雄激素分泌功能;其作用机制可能是通过下调CYP17基因及AR的表达实现的。结论:隐丹参酮可能具有潜在的治疗PCOS疾病的作用,值得进一步深入研究。
Objective: Through studying the effect of cryptotanshinone on hyperandrogenism of polycystic ovary syndrome (PCOS) and possible mechanisms to provide scientific basis for its clinical application. Methods: (~)Study on the effect of cryptotanshinone on reproductive endocrine function in hyperandrogenism model rats in vivo; (~)Study on the influence of cryptotanshinone on androgen secretion in hyperandrogenism model cell strain in vitro; (~)Study on the possible mechanisms of cryptotanshinone on hyperandrogenism of PCOS by detecting the expression of relvant proteins and the coded genes with irnmunohistochemistry (IHC) and real-time PCR. Results: After cryptotanshinone intervention, the estrous cyclicity, body weight, ovarian coefficient and ovarian morphology of PCOS model rats had been improved and the serum levels of testosterone (T), androstenedione (A2), luteinizing hormone (LH), sexual binding globulin (SHBG) had been reversed; The secretion of T of PCOS model cell had been suppressed. The possible regulatory mechanisms of cryptotanshinone on improving hyperandrogenism of PCOS might be related with down-regulating the expression of gene CYP17 and AR. Conclusion: Cryptotanshinone may have potential therapeutic effect on hyperandro~enism of PCOS oatients and deserves fi~rther inve.~ti~,atinn
