健脾理气法下调大鼠肝癌信号转导有关基因转录水平的筛选

目的:筛选出健脾理气法明显下调大鼠肝癌组织中与信号转导有关的基因。方法:以DEN诱导大鼠肝癌形成为模型,以西药喃氟啶化疗、中医清热解毒、活血化瘀治法为对照治疗,采用基因芯片技术,观察健脾理气法对大鼠肝癌组织基因组转录水平及信号转导相关基因转录的调控作用。结果:①与模型组比较,各中医治法下调最明显的基因分别为:清热解毒法明显下调140个基因,活血化瘀法明显下调186个基因,健脾理气法明显下调689个基因;②在健脾理气法明显下调的163个已知基因中,筛选出19个与信号转导相关的基因。讨论:基因芯片很好地揭示了不同中医治法各自防治肝癌的分子作用靶点,其中,健脾理气法下调大鼠肝癌组织与信号转导有关的基因转录中,涉及到细胞信号转导网络的多条通路的多个环节。

英文摘要:

Objective:The purpose of this study was to screen the signal transduction genes in rat liver - cancer tissues downregulated by invigorating spleen and regulating qi. Design:Induced by administration of DEN solution,we got the rat liver cancerg model to observe the regulation effects of invigorating spleen and regulating qi to genetic transcription, which is relevant to the transcription and signal transduction in genome of rats" liver - cancer tissues, controlled treatments about chemotherapy of medicine-Ftorafurum, and the two TCM treatments including expelling toxin by cooling and promoting blood circulation to remove blood stasis. Results:① compared to the model group,689 genes were down-regulated obviously by the treatment of invigorating spleen and regulating qi, while 140 genes by expelling toxin by cooling, and 186 genes by promoting blood circulation. ② screen 19 genes correlated with signal transduction among 163 known ones down - regulated by invigorating spleen and regulating qi. Conclusion:The genetic chips revealed that the method of down - regulated the genes associated with its signal transduction of liver cancer's tissues, referring toplenty of links in the lattice of transduction, among the functional molecule targets of these different treatment of TCM to prevent and cure liver cancer.
