目的研究p120-catenin(p120ctn)对肺癌细胞是否有调节作用,探讨其对细胞周期是否具有调节功能。方法应用siRNA方法建立p120ctn基因沉默的多种癌细胞系BE1,SPC和LTE的细胞克隆后,采用流式细胞术和裸鼠移植实验在活体内外观察p120ctn对肺癌细胞的生长是否具有调节作用。结果我们发现在p120ctn基因沉默的肺癌细胞系中,细胞的S期峰值明显增高,而G1期峰值明显减低。活体内的实验结果也显示注射p120ctn-siRNA肺癌细胞系的裸鼠形成的移植瘤的平均重量明显大于对照组和未转染组。结论 p120ctn参与细胞周期调节,缺失可促进肺癌细胞的生长。
Objective To study the effect of p120-catenin (p120ctn) on lung cancer cells to explore if p120ctn has regulatory role on cell cycle. Methods To investigate the role of p120ctn on cell cycle in lung cancer, we knocked down p120ctn using siRNA and measured the cell cycle with flow cytometer(FCM) analysis in vitro and nude mice transplantation in vivo. Results p120ctn knockdown groups showed a significant reduction in the percentage of G1 cells and a significant increase in the percentage of S phase cells in comparison to the untransfected cells and the negative control in vitro. The mean tumor weight in the p120ctn-siRNA group was significantly higher than that in either the untransfected group or control transfectant group in vivo. Conclusion p120ctn participated in the regulation of cell cycle, and p120ctn depletion promoted tumor cell mitosis and growth.
