免疫分析(Immunoassay,IA)是利用抗原(Antigen,Ag)和抗体(Antibody,Ab)之间的特异性反应来选择性识别和测定作为抗原或抗体的待测物.毛细管电泳免疫化学发光分析(Capillary electrophoresis immunoassay chemiluminescence detection,CEIACL)结合了CE的高分离效率、IA的高特异性及CL检测的高灵敏度的优点,同时还具有样品耗量少和仪器简单等特点.Mangru和Harrison首次利用辣根过氧化物酶(Horseradish peroxidase,HRP)对鲁米诺-过氧化氢的催化作用,应用芯片毛细管电泳分离化学发光法检测了HRP标记的羊抗鼠免疫球蛋白(IgG)的F(ab’)2片段和鼠IgG的免疫反应产物.Tsukagoshi等利用CEIACL检测了鼠IgG和HRP标记的抗鼠IgG免疫反应产物.本组利用CEIACL方法检测了鼠血管平滑肌细胞中骨形成蛋白-2,检出限达到6.24×10^-12mol/L.最近,Wang和Ren也开展了CEIACL研究,并实现了人血清中CA125的定量分析.
A novel detection method of a tumor marker alpha-fetoprotein (AFP) in human sera via capillary electrophoresis immunoassay coupled with chemiluminescence detection (CEIACL) was established. After a simple non-competitive immunoassay procedure, seras were directly injected into capillary for electrophoresis with chemiluminescence reaction of luminol and H2O2 catalyzed by horseradish peroxidase and enhanced by p-iodophenol. The detection limit (S/N = 3) of AFP is 0. 85 μg/L. The CEIACL method proposed was successfully applied to the sensitive detection of AFP in human sera, which coincides with the results detected by Beckman automatic CL analyzer-access IA system.
