用离子交换柱分离氨基糖苷类抗生素妥布霉素、新霉素B和西梭霉素,用脉冲积分安培电化学检测器进行检测。其流动相不同于一般抗生素的离子对色谱法,它配制简单,仅需使用NaOH-水体系;对两种检测电位波形进行了对比,并选取了表现优良的检测电位。方法对妥布霉素、新霉素B和西梭霉素的检出限分别为8、5、47ng/mL,在0.1~100/μg/mL范围内表现良好线性关系(r=0.9990~0.9996)。该方法应用于市售鲜奶的检测,3种氨基糖苷类抗生素古白回收率为850%~985%.
A new method was developed for detection of aminoglycoside antibiotics of tobramycin, neomycin and sisomyein by ion-exchange column with pulsed integrated amperometrie detection. The method was simple enough with the mobile phase of NaOH/H2O and a good detect waveform based on experiments. The detection limits for tobramyein, neomycin and sisomyein were 8, 5, 47 ng/mL respectively ; the linear range was 0.1 - 100 μg/mL ( r = 0. 9990 0. 9995). The method was applied to the detection of milk samples and spiked samples with recoveries of 85.0 % 98.5 % for the three antibiotics.
