目的研究美洲大蠊提取物(PAE)对口腔溃疡大鼠模型的作用。方法取大鼠50只,按照体重随机分为6组:空白组、模型组、阳性药组(3%桂林西瓜霜溶液)和3个剂量实验组(PAE,6%,3%,1.5%),苯酚溶液制备大鼠口腔溃疡模型,给药5 d,每天2次,每次1 mL。用流式细胞术检测外周血中T淋巴细胞亚群CD3+、CD4+、CD8+、CD4+/CD8+的变化情况,用酶联免疫吸附法(ELISA)检测血清中白细胞介素2(IL-2)水平,并观察口腔溃疡组织的病理变化。结果苯酚溶液可成功复制大鼠口腔溃疡模型。与模型组相比,阳性药组与高、中2个剂量实验组,口腔溃疡炎症均明显减轻,外周血中CD3+、CD4+细胞数和CD4+/CD8+比值显著降低(P〈0.05),而CD8+细胞数显著升高(P〈0.05);与模型组相比,阳性药组和3个剂量实验组,血清中IL-2水平明显升高(P〈0.01)。结论美洲大蠊提取物可降低机体的炎症因子水平和调节免疫功能,从而加快大鼠口腔黏膜愈合。
Objective To investigate the effect of Periplaneta americana extract( PAE) on oral ulcer in rat. Methods Fifty rats were randomized into 6 groups: blank group,model group,positive group( Guilin watermelon frost,3%),and three dose test groups( PAE,6%,3%,1. 5%). Rats in model with oral ulcer was induced by burning cheeks with phenol solution,one m L each time,twice a day,five days. T lymphocyte subsets CD3+,CD4+,CD8+,CD4+/ CD8+ in the peripheral blood were tested by flow cytometry. The level of interleukin 2( IL- 2)in serum was determined by enzyme linked immunosorbent assay( ELISA) and the change of pathological tissue was observed. Results The phenol solution was successfully used to establish the model of oral ulceration. Compared to the model group,the symptom of oral ulcer inflammation in positive group and the high and medium dosage test groups reduced significantly( P〈 0. 05). The cells of CD3+ and CD4+ and the ratio of CD4+/ CD8+ in peripheral blood in positive group and three dosage test groups also decreased significantly( P〈 0. 05),while the cells of CD8+ increased( P〈 0. 05). Compared to the model group,the IL-2 in serum in positive group and three dose test groups increased( P〈 0. 01). Conclusion The PAE can reduce the level of inflammatory factor and regulate the immune function,and thus can accelerate the healing of rats' mucosa.
