本文研究表面活性剂(MES,methyl ester sulfonate)、真空渗入法和二次侵染对提高花生转化效率的影响,建立了一种农杆菌介导的花生快速高效遗传转化体系。该体系以带子叶的胚轴为外植体,在OD600为0.7的菌液中,加入150mg/L的MES,40kPa真空处理10min,共培养3d后进行二次侵染。采用该体系可显著提高遗传转化率,GUS基因阳性表达率最高,达73.3%。
Impact of surfactant ( MES,Methyl ester sulfonate) ,vacuum infiltration and twice infection treat-ments on peanut ( Arachis hypogaea L. ) trans-efficiency was studied. The object was to construct a high efficient Agrobacterium tumefaciens-mediated transformation system. The explant of hypocotyl with cotyledons were cultured in the pMMO which value in OD600was 0. 7,adding the MES in a concentration of 150mg/L,and treated in vacuum at 40kPa for 10 minutes. The explant was infected for the second time after being cultured for 3 days. This system could apparently improve the efficiency of transformation,in which the GUS gene could be expressed as high as 73. 3% .
