应用超声破碎法分别在HCl(0.01mol/L)、蒸馏水及NaOH(4%)3种不同介质中提取自养小球藻(Chlorella autotropica)多糖。经3%TCA除蛋白及色素、乙醇沉淀真空干燥后,采用二乙胺基乙基琼脂糖凝胶柱层析。实验结果表明,以NaOH(4%)为介质进行超声破碎法提取并层析纯化是一种较适合自养小球藻多糖提取纯化的方法。
The ultrasonic crushing method was used to extract the polysaccharide from Chlorella autotropica in three media of HCI(0.01 mol/L), distilled water and NaOH(4 %). After removal of protein and pigment from polysaccharide with 3 % TCA, ethanol precipitation and vacuum desiccation, the DEAE sepharose column chromatographic analysis of purified polysaccharide was made. It is shown from the analysis results that the method using ultrasonic crushing technique, extraction of polysaccharide in NaON(4 %) medium, removal of protein and pigment with 3% TCA and purification of poiysaccharide with DEAE sepharose column chromatography is a suitable extraction and purification method of polysaccharide from Chlorella autotropica.
