CD59-siRNA对人卵巢癌细胞裸鼠移植瘤的作用

目的探讨针对CD59的siRNA(CD59-siRNA)对人卵巢癌裸鼠移植瘤CD59的沉默效应及其抑瘤作用。方法采用脂质体转染法将CD59干扰质粒(T组)和空质粒(V组)转染A2780细胞,获得稳定表达细胞株,将T组、V组A2780细胞和未转染(C组)的A2780细胞分别接种于裸鼠皮下建立肿瘤模型,通过绘制肿瘤生长曲线、RT-PCR和免疫组织化学法研究其抑瘤效应和对CD59基因及蛋白的沉默效应。结果与V组和C组比较,T组肿瘤体积和质量明显减小(F=301.88、5.39,q=4.01~30.41,P〈0.05),CD59基因及蛋白表达减少(F=817.77、247.71,q=26.59~52.25,P〈0.05)。结论特异性沉默CD59基因的siRNA表达载体可以明显抑制CD59的表达及卵巢癌在体内的生长。

Objective To investigate the inhibition effect of CD59-siRNA on CD59 gene and the growth of ovarian cancer in nude mice. Methods A2780 cells were stably transfected with shRNA plasmid(group T) and blank plasmid(group V),respectively,using Lipofectamne 2000.These cells and A2780 cells without trnsfection(group C) were subcutaneously injected into nude mice.The inhibition effect of CD59-siRNA on tumor and the expreesion of CD59 was evaluated by tumor growth curve,RT-PCR and immunohistochemistry(IHC). Results Compared with groups V and C,the gross tumor volume and weight were significantly inhibited in group T(F=301.88,5.39;q=4.01-30.41;P0.05),RT-PCR and IHC showed that the expression of CD59 mRNA and CD59 also decreased(F=817.77,247.71;q=26.59-52.25;P0.05). Conclusion CD59-siRNA can significantly suppress the expression of CD59 and growth of ovarian cancer in vivo.
