以油松外生菌根真菌球根白丝膜菌(Leucocortinarius bulbiger(Alb.et Schw.)Sing.)的子实体为材料,采用组织分离方法获得纯菌种,接种于不同培养基,筛选出最适基础培养基.运用单因素优化法筛选出最佳碳源、氮源、pH值,再经多因素正交试验筛选出菌丝体生长最佳碳源、氮源及pH值组合.结果表明,球根白丝膜菌菌丝体生长最适基础培养基为PACH,在此基础上筛选的菌丝体生长最佳碳源、氮源、pH值分别为葡萄糖、酒石酸铵、pH=7.0.正交试验筛选的菌丝体生长的最快三因素组合为蔗糖、磷酸二氢铵、pH值为7.0.
Fruiting bodies of Leucocortinarius bulbiger which is Pinus tabulaeformis' Ectomycorrhizal fungi were taken as research material,pure strain was obtained using tissue isolation method.The strain was inoculated in different culture medium,optimum culture medium was selected.The best carbon source,nitrogen source and pH value were selected using the single factor optimization method.The optimum combination of carbon source,nitrogen source and pH value was selected by using multi factor orthogonal experiment.The results indicated that Leucocortinarius bulbiger's best basic culture medium was PACH.On the basis of above result,the optimum carbon source,nitrogen source and pH value of mycelium growing were respectively glucose,ammonium tartrate and pH7.0.Using orthogonal experiments,the three factor combination selected which lead to mycelium growing fast was sucrose,ammonium dihydrogen phosphate and pH 7.0.
