目的 探讨糖氧剥夺(oxygen-glucose deprivation,OGD)对体外培养的大鼠星形胶质细胞窖蛋白1(caveolin-1,Cav-1)的影响.方法 原代培养出生24 h内SD大鼠大脑皮质星形胶质细胞,分为对照组和OGD组(给予星形胶质细胞6h的OGD和24h的再灌注损伤,建立OGD损伤模型).用多重免疫荧光的方法检测星形胶质细胞Cav-1表达,实时荧光定量PCR和Western blot的方法检测星形胶质细胞Cav-1表达的变化.根据小干扰RNA(siRNA)的原理,将Cav-1 siRNA转染星形胶质细胞,采用CCK-8的方法检测星形胶质细胞活力.结果 大鼠大脑皮质星形胶质细胞Cav-1普遍表达.与对照组比较,OGD组星形胶质细胞Cav-1 mRNA和蛋白表达量下降(P<0.05).siRNA转染后,与对照组比较,OGD组星形胶质细胞活力下降(P<0.05).结论 SD大鼠大脑皮质星形胶质细胞Cav-1表达的下调,导致OGD后细胞损伤加重,提示Cav-1对星形胶质细胞具有保护作用.
Objective To study the effect of caveolin-1 (Cav-1) on oxygen glucose deprivation (OGD)-induced injury of in vitro cultured astrocytes in rats.Methods Cerebral cortex astrocytes from 1 day old SD rats were primarily cultured.An OGD model was established.The cells were divided into control group and OGD group.The cells in OGD group were subjected to OGD for 6 h and reperfusion for 24 h.Expression of Cav-1 in astrocytes was detected by immunofluorescence,RT-PCR and Western blot,respectively.Cav-1 siRNA was transfected into astrocytes.The survival rate of astrocytes was assayed with the CCK-8 method.Results The Cav-1 was expressed in astrocytes.The expression level of Cav-1 in astrocytes was significantly lower in OGD group than in control group after siRNA transfection (P〈0.05).Conclusion Down-regulated Cav-1 expression aggravates OGD-induced astrocyte injury in rats,indicating that Cav-1 can protect astrocytes against OGD-induced injury.
