目的 建立经济简便的小鼠可卡因自给药模型,研究β1肾上腺素受体拮抗剂倍他洛尔和β2肾上腺素受体拮抗剂ICI 118,551对小鼠可卡因自给药维持的影响。方法 对小鼠静脉插管手术和自给药训练的方法进行改良,建立小鼠可卡因自给药模型。在小鼠形成稳定自给药行为后,分别腹腔注射非选择性的β肾上腺素受体拮抗剂普萘洛尔(10 mg/kg)、受体亚型选择性的拮抗剂倍他洛尔(10 mg/kg)或ICI 118,551(10 mg/kg),观察3种药物对小鼠固定比率自身给药行为维持的影响。结果 成功建立小鼠固定比率自给药行为。给予普萘洛尔组和ICI 118,551组小鼠固定比率给药次数明显降低,倍他洛尔组小鼠固定比率给药次数无明显变化。结论 ICI 118,551可降低小鼠可卡因自身给药行为,而倍他洛尔对小鼠可卡因自身给药行为没有明显的影响。
Objective To establish an economic and simple behavioral model of intravenous cocaine self-administration in mice,and to investigate the effects of selective β1-adrenergic receptor antagonist betaxolol and selective β2-adrenergic receptor antagonist ICI 118,551 on the maintenance of drug selfadministration in mice.Methods Cocaine self-administration was established under an FR1 reinforcement schedule with our modified method of intravenous catheterization and self-administration training.Once mice had established stable cocaine self-administration behavior,they received intraperitoneal injection of nonselective β-adrenergic receptor antagonist propranolol (10 mg/kg),β1-antagonist betaxolol (10 mg/kg),or β2-antagonist ICI 118,551 (10 mg/kg).The effects of the drugs on the maintenance of cocaine self-administration were evaluated.Results Mice successfully acquired cocaine self-administration under an FR1 reinforcement schedule.Intraperitoneal administration of propranolol or ICI 118,551 reduces in cocaine selfadministration,whereas pretreatment with betaxolol made no significant changes.Conclusions β2-adrenergic receptor antagonist ICI 118,551 could decrease cocaine selfadministration in mice,whereas β1-adrenergic receptor antagonist betaxolol had no significant effect on cocaine selfadministration.
