目的:观察中波紫外线对人血小板线粒体活性及线粒体DNA的影响,探讨其作为评测光损伤敏感系统的可行性。方法:以不同能量的中波紫外线照射人血小板,观察血小板线粒体活性及线粒体DNA光化产物环丁烷嘧啶二聚体(CPDs)含量的变化。结果:接受不同能量中波紫外线照射的血小板,线粒体活性明显降低(P〈O.05),环丁烷嘧啶二聚体(CPDs)含量则明显增加(P〈0.05)。结论:人血小板线粒体活性与环丁烷嘧啶二聚体含量对中波紫外线的照射敏感,并与其剂量呈依赖关系,可很好反映中波紫外线对生物体光损伤作用。
Objective To investigate whether human platelets could be applied on photoaging research. Methods After human platelets were irradiated with different doses of ultroviolet B, changes of mitochondria activity and cyclobutane pyrimidine dimmers (CPDs)in mitochondria DNA were observed. Results Change of mitochondria activity had significant difference(P〈0.05), and content of CPDs in mitochondria DNA increased with doses of UVB. differences were statistical significance (P〈0.05). Conclusion Preliminary study deduce that mitochondria activity and content of CPDs in mitochondria DNA were sensitive to different doses of UVB, and human platelets can be applied on photic injury research.
