摘要:以SD大鼠为研究对象,研究高脂饮食诱导肥胖(DIO)和肥胖抵抗(DDIO—R)模型大鼠组织中水通道蛋白(AQP)7和9mRNA的表达情况,探讨其在肥胖发生中的作用机制。将38只雄性SD大鼠随机分为正常对照组(CK组,n=8)和造模组,分别饲喂基础饲料和高脂饲料。8周末,造模组按体重筛选出D10组和DIoR组大鼠。20周后处死,计算Lee’s指数和脂体比,检测血清生化指标,Real-timePCR测定AQP7和AQP9mRNA表达。结果表明:DIO组大鼠体重、脂体比、三酰甘油(TG)、血清胰岛素(FINS)和胰岛素抵抗指数(HOMA-IR)均显著高于CK组和DIO-R组,Lee’s指数和总胆固醇(TC)显著高于CK组,DIO-R组大鼠脂体比、FINS和HOM小IR也显著高于CK组;DIO组和DIO-R组大鼠肾周脂肪AQP7mRNA表达显著下调;DIO组大鼠骨骼肌AQP7和肝脏AQP9mRNA表达比CK组和DIO-R组显著降低。可见不同肥胖易感大鼠肾周脂肪AQP7mRNA表达水平一致,骨骼肌AQP7和肝脏AQP9mRNA表达水平存在差异。
ABSTRACT: To observe tissue expression of aquaporin 7 (AQP7) and aquaporin 9 (AQP9) mRNA between high-fat diet induced obese rats (DIO group) and obesity resistant rats (DIO-R group) and investigate the related mechanism in obesity development, 38 male Sprague-Dawley rats were randomly divided into control group (n=8) and model group which fed with. normal and high-fat diet, respectively. Rats in model group were subdivided into DIO group and DIO-R group ac- cording to body weight at 8th week. The rats were all sacrificed at 20th week. Lee's index, fat content, serum biochemi- cal indices were determined. AQP7 and AQP9 mRNA expression were measured by Real-time PCR. The results showed that body weight, fat content, serum TG, FINS and HOMA-IR in DIO group were higher than those in control and DIO- R group (P〈0. 05), Lee's index and serum TC level were higher than control group (P〈0.05), while rats in DIO-R group also showed higher fat content, serum FINS and HOMA-IR comparing with control group (P〈0.05). AQP7 mR- NA in perirenal adipose of DIO and DIO-R group were lowed than those in control group (P〈0.05). Expression of AQP7 mRNA in skeletal muscle and AQP9 mRNA of DIO group were lower than control group and DIO-R group (P〈 0.05). These data suggested that among rats with different susceptibility to obesity, AQP7 mRNA in perirenal adipose showed same expression, but AQP7 mRNA in skeletal muscle and AQP9 mRNA in liver indicated differential expression.
