研究旨在探讨丙酮酸激酶M1(pyruvate kinase M1,PKMl)、丙酮酸激酶M2(pyruvate kinase M2,PKM2)在不同结肠癌细胞中表达情况与结肠癌发展的相关性。通过Westernblot、实时荧光定量PCR、丙酮酸激酶活性分析,检测不同恶性程度结肠癌细胞中PKMl、PKM2的蛋白与基因表达水平及丙酮酸激酶活性,分析它们与结肠癌恶性程度的相关性。PKMl和PKM2在人结肠癌细胞和正常结肠上皮细胞中均有表达;PKMl及PKM2的总蛋白和mRNA表达情况与人结肠癌恶性程度无明显相关性;细胞核中PKMl与PKM2的表达量比值与结肠癌恶性程度呈负相关;丙酮酸激酶活性与结肠癌恶性程度呈正相关。细胞核中PKMl与PKM2的表达量比值可以作为结肠癌恶性评价的一个指标。
This study detects the expression of PKM1 and PKM2 in different malignant colon cancer cell lines through detecting gene transcription and protein expression levels. The correlation between the pyru- vate kinases and colon cancer development was studied. Real-time PCR is used to determine the expression levels of gene PKM1 and PKM2. Western blot is applied to detect the expression of PKM1 and PKM2 pro- tein. The pyruvate kinase activity was analyzed in human colorectal cancer cells. The results indicate that there are PKM1 and PKM2 expression in both colon cancer cells and normal colon epithelial cells. Howev- er,there is no clear correlation between either PKM1 and PKM2 mRNA or total protein expression with various stages of colon cancer cells. Notably,the ratio of PKM1 expression and PKM2 expression in cellular nucleus is associated with colon cancer negatively. In addition,Pyruvate kinase activity is positively correla- ted with malignant degree of colon cancer. The ratio of PKM1/PKM2 is correlated with the malignant de- gree of colon cancer strongly and it might be use as a tumor therapeutic index in the future.
