目的 将xMAP(flexible multi—analyte profiling)技术和ELISA(enzyme—linked immunosorbent assay)应用到人泪液中的表皮生长因子(epidermal growth factor,EGF)检测中,探讨xMAP技术在泪液检测中的应用前景。方法 选取24例正常人(年龄18~33岁),在裂隙灯下取无刺激性泪液,用xMAP法检测泪液中EGF的浓度,与ELISA测定的结果比较,进行统计学分析。结果 xMAP法检测泪液中EGF浓度为(4.32±2.57)ng/ml(范围0.96~9.76ng/ml),同一批样本经ELISA检测为(8.78±5.35)ng/ml(范围2.45~20.39ng/ml),两种方法的结果相关系数R^2=0.8508(p=0.000)。结论 xMAP法与ELISA法相关性良好,可以用于泪液中多种微量蛋白的检测。
Objective To investigate the application of xMAP (flexible multi-analyte profiling) technology compared to enzyme-linked immunosorbent assay (ELISA) in the assay of EGF in human tear. Methods Non-reflex tear from twenty-four healthy humans were collected under slit-lamp biomicroscope. The EGF concentration in human tear was measured by xMAP. Meanwhile, the ELISA method was used for comparison. All data was analyzed by the statistic software SPSS. Results The EGF concentration determined by xMAP was (4.32 ± 2.57)ng/ml (range: 0.96-9.76ng/ml) in humans (age: 18-33 yrs). It was 8.78 ± 5.35ng/ ml (range: 2.45-20.39ng/ml) when measured by ELISA. The correlation coefficient (R^2) of the data assayed by two methods was 0.8508 (p=0.000). Conclusion There is a good relation between xMAP and ELISA in the determination of the EGF in human tear. The xMAP technology can be used in the assay of trace proteins in human tear.
