目的:对快速老化模型小白鼠SAMP8老化征象和学习记忆能力的增龄性变化进行系统研究,为利用该模型进行其它研究提供实验依据。方法:选用2、4、6、8、10、12月龄SAMP8,以同龄正常老化的SAMR1为对照,应用老化度评分和Morris水迷宫测试SAMP8的老化度和学习记忆能力的增龄性变化。结果:SAMP8在4月龄出现明显的老化体征,并在6月龄开始表现出空间学习记忆障碍。结论:SAMP8在成熟期(4-6月)过后发生了快速老化,证实SAMP8是研究衰老和老年性痴呆的理想模型。
Objective: To observe the age-related changes in aging sign and symptoms, and learning and memory of senescence accelerated mouse SAMP8. Methods: Senescence Accelerated Mouse SAMP8 and normal control SAMR1 at six ages from 2th month to 12th month were selected, and age-related changes in aging and learning and memory were studied by " the scores of aging" and Water Morrris Maze. Results: The SAMP8 appeared obvious aging sign and symptoms at 4-month old. Compared to SAMR1 control group, SAMP8 mouse showed significant impairments on learning and memory in hidden platform trial at age of 8th month, 10th month and 12th month. In probe trial and reversal trial, SAMP8 began to show significant impairments on learning and memory at age of 6month. Conclusions: The results showed that SAMP8 began to accelerate senescence after mature period (4-6month) and appeared learning and memory disorder at 6-8 months old. It manifested that SAMP8 was an ideal model for investigation on the mechanism of aging and senile dementia.
