目的探讨激活素A对小鼠腹腔巨噬细胞分泌细胞因子的影响。方法分离小鼠腹腔巨噬细胞,将其分为激活素A处理组和对照组,瑞氏-吉姆萨染色观察小鼠腹腔巨噬细胞形态学变化;流式细胞术分析小鼠腹腔巨噬细胞表面分子CD68的表达;ELISA法检测小鼠腹腔巨噬细胞分泌IL-10及TNFα的水平;Griess法检测小鼠腹腔巨噬细胞分泌NO的水平。结果经激活素A刺激后,显微镜下可见呈不规则、多边型的活化巨噬细胞增多,细胞表达巨噬细胞成熟标志CD68增加,Ⅱ型巨噬细胞(M2)产生的细胞因子IL-10及NO分泌水平升高,而Ⅰ型巨噬细胞(M1)产生的细胞因子TNFα水平无变化。结论激活素A可能主要促进小鼠Ⅱ型巨噬细胞分泌细胞因子。
Objective To investigate the effect of activin A on cytokine secretion from peritoneal macrophages of mice. Methods The peritoneal macrophages of mice were isolated and divided into two groups. The peritoneal macrophages in test group were cultured in the RPMI1640 medium containing activin A, while those in control group in the RPMI1640 medium containing no activin A. The peritoneal macrophages were observed for morphology by Wright-Giemsa staining, analyzed for expression of surface molecule CD68 by flow cytometry, and determined for secreted IL-1 and TNFα levels by ELISA and for secreted nitrogen oxide level by Griess method. Results After stimulation with activin A, irregular and polygonal activated mouse peritoneal macrophages in an increased number were observed by microscopy, while the expression of CD68 was upregulated, and the IL-10 and nitrogen oxide lev-els secreted by macrophages type Ⅱ(M2)increased. However, the TNFα level secreted by macrophages type Ⅰ(M1)showed no significant change. Conclusion Activin may mainly promote the secretion of cytokines by M2.
