目的制备一种以卡波姆(carbomer)胶为基质的载体用于黑素细胞涂布法移植。方法体外培养正常人的黑素细胞,配制卡波姆胶半固体培养基;分别采用台盼蓝染色、噻唑蓝比色法[3-(4,5-Dimethyl-2-Thiazolyl)-2,5-Diphenyl Tetrazolium Bromide,MTT]检测1%卡波姆胶对黑素细胞毒性和增殖的影响;用Transwell培养板检测1%卡波姆胶对黑素细胞迁移的影响。结果 1%卡波姆胶作用0~36 h对黑素细胞活性和增殖无影响;1%卡波姆胶作用12 h后对黑素细胞迁移无影响(P〉0.05)。结论采用卡波姆胶混合黑素细胞,不影响体外培养的黑素细胞功能,可以作为新型黑素细胞涂布法移植载体。
Objective To prepare a carrier with carbomer gel as substrate for melanocytes transplantation by spreading method.Methods Normal human melanocytes were cultured in vitro.Then,a semi solid medium with carbomer gel was prepared as substrate.The toxicity and proliferation of the melanocytes were tested by Trypan blue and 3-(4,5-Dimethyl-2-Thiazolyl)-2,5-Diphenyl Tetrazolium Bromide(MTT).Transwell culture plates were employed to assay the migration of melanocytes.Results 1% carbomer gel had no negative impact on the activities and proliferations of melanocvtes(P0.05).The carbomer gel didn't inhibit the migration of melanocyte after 12 h(P0.05).Conclusion The mixture of carbomer gel does not injure the biological function of melanocytes,which is suit for preparing the carrier for melanocyte transplantation.
