采用光镜和透射电镜技术,探讨了不同浓度纳米氧化锌暴露对鲤鱼幼鱼鳃显微和超微结构的影响。暴露组纳米氧化锌浓度分别为5、10、20、50、100 mg.L-1,同时设对照组,连续暴露21 d,取鳃制备切片,进行光镜和透射电镜观察。结果表明,21 d后纳米氧化锌暴露组鲤鱼鱼鳃部均有不同程度的损伤,石蜡切片上可观察到暴露组鱼鳃上皮细胞增生、变性、鳃小片断裂或融合,且病变程度随纳米氧化锌浓度的升高而加重;透射电镜下可观察到暴露组鱼鳃氯细胞内线粒体出现肿胀、嵴模糊或缺失、空泡化。表明纳米氧化锌暴露可在短期内对鲤鱼鱼鳃组织造成损伤,且损伤程度存在剂量依赖效应。
The effects of Nano-ZnO on microstructure and ultrastructure of the gill of Carp(Cyprinus carpio) were estimated by optical and transmission electron microscopy.Results showed that gills from Carp exposed in Nano-ZnO at the concentration of 5,10,20,50,and 100 mg·L-1 for 21 days had obvious structural alteration.The histopathological changes included hyperplasia and degeneration of epithelia cells,rupture and malgamation of gill lamellae,which became severer with the increase of Nano-ZnO concentration.Ultrastructural changes showed that mitochondria in chloride cell appeared tumefaction with blu,deletion of cristae,and vacuolization.The results indicated that Nano-ZnO could cause damage to the gill of Carp after short period exposure,and damage degree had the dose-dependent effect.
