为了筛选最适合于图谱构建的多态性比率高、扩增效果好的分子标记技术,以吉林省露水河林业局红松种子园某家系中的父母本和部分子代为研究对象,通过多态性的检测效率等指标比较了SSR、ISSR和SRAP 3种分子标记方法应用于红松遗传图谱构建的可行性。结果表明,SRAP分子标记检测作图亲本间多态性的效率最高,为3.06;其次是SSR标记,多态性检测效率为2.67;ISSR标记最低,为1.5。结合3种分子标记方法的经济成本、技术要求等因素,得出了结论:在构建红松图谱时应以SRAP为主要分子标记技术,SSR标记和ISSR标记为辅助标记,更为经济、高效。
In order to explore the suitable molecular marker technique which had the most efficient polymorphism detection efficiency and the best amplification effect in map construction of Pinus Koraiensis,SRAP,SSR and ISSR markers were performed in parents of a Korean pine clone and some of their F1 progeny collected from Korean Pine Seeds Park in Forestry Bureau of Lushuihe,Jilin Province.The results showed that SRAP was the most efficient and the detection efficiency was 3.06;SSR was the second and the detection efficiency was 2.67;ISSR was the least efficient and the detection efficiency was 1.5.Combined with the economic costs,technical requirements and other factors,it was concluded that SRAP should be the main molecular marker in map construction of Korean pine,SSR and ISSR were complementary molecular markers which were more economical and efficient.
