深Ⅱ度烫伤早期犬血液生理指标及四种细胞因子质量浓度的动态变化

以中华田园犬为研究对象,用99口C多层纱布覆盖犬腰荐部多个面积为3cm。区域皮肤,制备深Ⅱ度烫伤模型,采集烫伤前和烫伤后第6、12、24和48小时血样和烫伤皮肤组织样品,检测血液生理指标和细胞因子IL-6、IL-8、IL-10、TNF-α的质量浓度。结果显示,烫伤后犬血液白细胞、中性粒细胞和单核细胞的浓度均显著升高(P〈0.05),烫伤后第24小时达到峰值,淋巴细胞的浓度显著降低(P〈0.05),烫伤后第12小时降至最低;犬血清和组织中IL-6、IL-10和TNF-α的质量浓度在烫伤后第6~24小时均显著高于烫伤前(P〈0.05),烫伤前后血清中IL-8的质量浓度未发生显著变化(P〉0.05),组织中IL-8的质量浓度烫伤后第24小时显著升高(P〈0.05)。结果表明,深Ⅱ度烫伤早期(48h以内)试验犬呈现全身性炎性反应特征,血清和烫伤组织中促炎/抗炎细胞因子水平不同程度升高。

英文摘要:

The present study aimed to observe the dynamic changes of blood physiologic index and cy- tokines in serum and scalded tissue of dogs at the early stage of deep partial-thickness burn. Chinese rural dogs were used to produce the deep partial-thickness burn model by multilayer pledgets at 99℃ overlap- ping the lumbar-sacral skin with 3 cm2 area for 23 s, thus the blood samples were taken at the different stage of pre- and at 6 h,12 h,24 h and 48 h post-scald,in order to detect blood physiology index with auto- matic Blood Cell Counter, and the levels of cytokines IL-6,IL-8,IL-10 and TNF-α in serum and scalded tissue with ELISA kits. In result, the counts of WBC,GRA and MID significantly increased in post-scalded canine blood compared with that in the pre-scald canine blood(P〈0.05) ,and reached the maximum at 24 h post-scald,while LYM's counts significantly decreased(P〈0.05) and reached at the minimum at 12 h post-scald. In the experimental period from 6 h to 24 h post-scald, the levels of IL-6, IL-10 and TNF-α sig- nificantly increased in both serum and tissue scalded(P〈0.05). The level of IL-8 in serum showed no sig- nificant difference between pre- and post-scald(P〉0.05),but the level in the tissues was significantly higher 24 h post-scald than those at other time points (P 〈 0. 05). These results suggested that the experimental dogs presented characterization of systemic inflammatory response at the early stage of deep partial-thickness burn(within 48 h), and the pro-/anti-inflammatory cytokines increased to different extents in serum and the scalded tissues.
